Alkaline and acid phosphatase activity in semen of Mantiqueira bulls

Abstract

The purpose of this paper was the determination of the phosphomonoesterase activity (alkaline and acid phosphatases) in hundred and sixty samples of seminal plasma from eight brazilian Mantiqueira bulls. Thus, the alkaline phosphatase activity were determined by the colorimetric method of KING & ARMSTRONG (1934) using phenyl phosphate disodium salt as substrat, incubated at 37°C for 15 minutes and the spectrophotoznetric lecture of the colorimetric product was made at 680 nm. The acid phosphatase activity were determined by the colorimetric method of FISHMAN & DAVIDSON (1957), using phenyl phosphate disodium salt as substrat, incubated at 37° for 60 minutes and the spectrophotometric lecture of the colorimetric product was made at 510 nm. The average obtained for alkaline and acid phosphatases were 467.75 ± 27.96 and 338.58 ± 39.58, respectively. The acid phosphatase presents positive correlation (P < 0.1) with alkaline phosphatase and negative (P < 0.05) with the volume of ejaculated of bovine seminal plasma. The results showed that the methods employed had good reproductibility and they may be used to the stablishment of the standards of enzimatic activities in plasma seminal of bulls used in artificial insemination.
